Solid Phase Synthesis Materials

Redstone Separations AB develops solid phase synthesis resins for various types of supported step-wise chemical reactions and synthesis. The most prominent application area is the prepration of API peptides and oligonucleotides up to a certain length. 

Conventional solid phase peptide support resins are polymeric materials with low levels of cross-linkage, which are the standard today for conventional solid phase peptide synthesis. As gel-type materials, these do not display distinct pore sizes but have certain swelling characteristics in various solvents.

Novel solid phase supports are highly cross-linked, with large pores aimed at implementation in oligonucleotide synthesis could also be envisaged for use in novel flow peptide synthesis or for other molecules that benefit from a rigid bead structure or from being used in a flow system.

Our current development portfolio has the following cross-linked polymeric resins in the pipeline:

  • DVB/Styrene reversed phase resins
  • X-link variations:
    • low cross-linkage (1 or 2 %)
    • high cross-linkage (eg 50 %)
  • Particle sizes:  20-45 µm / 45-70 µm / 70-200 µm or larger
  • Porosity:
    • gel-type
    • mesoporous 20-500 Å
    • macroporous 500 Å to 1000 Å
    • also >1000 Å (upon inquiry)
  • Capacity: 0.1 mmol to 2 mmol/g 
  • Other properties: upon request

A first study on Redstone SABIT SPPS prototypes was conducted in a master thesis project in collaboration with Copenhagen University, Denmark.

Abstract

Solid-phase peptide synthesis (SPPS) is the most widely used method for producing synthetic peptides, with the solid support-known as the resin-being a critical component of the process. Redstone Separations AB, a company based in Lomma, Sweden, has developed new SPPS resins and aims to evaluate their performance compared to commercially available alternatives.

In this study, Redstone ́s resins were assessed by synthesizing three peptides:
Neurotensin, GLP-1 (at both room temperature and elevated temperature), and an RGD-peptide. These peptides were synthesized using SPPS followed by post-synthetic modifications for the RGD peptide, and resulting products were evaluated for yield and purity.

The results showed that for Neurotensin, some Redstone ́s resins outperformed some commercial resins, although the highest performance was still observed with the commercial resins. In the synthesis of GLP-1, both at room and elevated temperatures, commercial resins yielded better results, but Redstone ́s resins were comparable. The synthesis of the RGD-peptide was unsuccessful for both Redstone and commercial resins, as the obtained amounts were too low to quantify and the right peptide was not seen for some of the resins.

Overall, the results demonstrate that Redstone ́s resins are capable of producing peptides in reasonable yields and purities. Although further optimization is needed to reach or surpass the performance of established commercial resins, these findings are promising, especially considering that Redstone is newly established maker of SPPS resins in the market and that it was shown that the resins worked for SPPS overall.
